Abstract:
Calcium-triggered intracellular signalling is prerequisite to mount plant stress responses, and increasing evidence also hints toward a role of Ca2+ -dependent regulation in the control of plant growth and development. Calcium-dependent protein kinases (CDPKs) combine within one molecule a calcium-sensing domain, which contains in general 4 EF-hand calcium-binding motifs, and a protein kinase effector domain. Accordingly, CDPKs function as decoders that sense and translate (induced) changes in intracellular Ca2+ into further downstream events.
In innate immune signalling, upon recognition of bacterial pathogens, CDPKs have been identified as positive as well as negative regulators for both local early and late systemic defence responses in the context of both, PAMP-triggered immunity and Effector-triggered immunity. In particular a role in defence signal propagation has been shown. Deduced from these data we have initiated analogous studies with respect to wounding and herbivory and identified members of the CDPK gene family that participate in early signal propagation and defence response activation in herbivory.

Abstract:
Pyrrolizidine alkaloids (PAs) are toxic secondary metabolites as part of the plants chemical defense against herbivores. PAs occur in several unrelated lineages of the angiosperms. Studying the first specific enzyme of this pathway, homospermidine synthase (HSS), it was possible to show that the ability to produce PAs evolved several times independently during angiosperm evolution. Therefore, the system of PA biosynthesis is a promising tool to study pathway evolution, as comparisons of the evolutionary scenario in the various plant lineages are possible. Studies on the evolution of HSS allowed conclusions about the mechanisms that gave rise to a new enzymatic activity and show that common models of gene evolution have to be adapted. In addition, this data show that the characteristic diversity of secondary metabolism does not only refer to the chemical diversity of Pas, but also to regulation. Only specific cells are suggested to be involved in PA biosynthesis, often only during a restricted time window of plant development. The high specificity of expression in time and space is used to enrich transcriptomic libraries in candidate sequences for elucidation of further steps of this pathway. The most recent results on the evolution of PA biosynthesis will be presented and discussed.
